This research work lays down the foundation of the understanding of efficiency, specificity, fidelity and efficacy of gene editing.• Hands-on experience in gene targeting to generate mouse genetic models (knock-in/knock-out/transgenic), which serve as a strong foundation in understanding any given mouse disease models that are often necessary needed for testing drug candidate in proof of concept studies. From 2006 to 2009 (3 years) Predoctoral Research Associate @ UNC • Extensive fundamental training in AAV biology, vectorology and gene delivery technology, which sets the technical foundation required for gene therapy drug development.• Research results uncovered the requirement of mammalian cells DNA recombination and repair pathway proteins ATM, NBS1 and DNA-PK in AAV DNA genome circularization and concatemerization. These molecular mechanisms directly determine the fate of rAAV vector DNA to persist as episomes in the nucleus of the cells, which in turn leads to durability of gene expression.• Served as a self-complementary rAAV expert to collaborate with investigators from UNC Chapel Hill, Duke University, New Jersey Medical School, Colorado State University and U of Pittsburgh and enabled them to utilize rAAV gene delivery technology to perform translational research for joint, glaucoma, renal and cardiac diseases.
