From April 2013 to June 2013 (3 months) San Francisco Bay AreaPostdoctoral Fellow @ UCSF Investigated neural circuits involved in innate behaviors in rodent models using molecular and genetic approaches From April 2012 to April 2013 (1 year 1 month) San Francisco Bay AreaDoctoral Student, Program in Developmental Biology @ Baylor College of Medicine Doctoral student in the laboratory of Dr. Richard KelleyWorked on elucidation of the epigenetic regulation of transcription elongation using the Drosophila dosage compensation modelProject: Identification and characterization of novel molecules involved in regulation of the dosage compensation complex, MSL complex.Approach: Adopted an interdisciplinary approach that combined unbiased forward genetics, molecular biology and biochemistry to identify and characterize factors that regulate MSL complex function in vivo. Designed a novel genetic strategy that used a simple visual read out of MSL complex activity and performed two forward genetic screens.Results: Identified 48 mutations and 11 deletions that lowered MSL complex activity. Mapped mutations in Spt5, Sin3A, Chromator and MSL1. Characterized the mechanistic role of mutations in Spt5 and MSL1. Identification of mutations in the conserved elongation factor Spt5 provides the first direct molecular link that validates the elongation model of dosage compensation.
